It is highly dependent upon the type of clothing, and how dense the fibers are weaved together. Cotton is one of the easier fibers for a flea to penetrate.

A layer of light clothing is usually a sufficient protection from flea bites, but not always.

You’re more likely to be bitten along areas of the skin where the clothing is stretched tight across the skin, such as the waistline.

In order to more fully protect from flea bites, wear clothing in which the fiber is dense and knitted tightly together. Synthetic fiber such as those found in hiking gear are especially suited for this.
